2D CRINEPT

DESCRIPTION
In the case of large systems, scalar couplings are suplemented or even supplanted by cross-correlated relaxation to achieve magnetization transfer. One example is the CRINEPT (Cross-correlated Relaxation Enhanced Polarization Transfer) pulse sequence.

This approach offers the posibility to extent the aplication of triple-resonance NMR experiments to biomolecules larger than 25-30 kDa.

REQUIREMENTS
Easy implementation on AVANCE spectrometers equipped with pulsed field gradients (PFGs) and inverse probehead.
VERSIONS
The CRINEPT scheme ( 99PNAS4918 and 00TIBS462 and 03BIO8579 ).

  • Comparison of HSQC, TROSY, TROSY-Kay and CRINEPT for very large proteins at 900MHz ( 03JMR360-163 ). See 2D TROSY experiment for more info.
  • CRIPT (Cross-correlated relaxation-indiced polarization transfer) experiment ( 02JACS12144 , 05JB23-32 ). Study on the effect to manage the solvent water magnetization to obtain improved CRIPT spectra ( 05JB61-32 ).
  • EXPERIMENTAL DETAILS
    This experiment can be recorded using the basic parameter set available for a 2D inverse-detected experiment as, for instance, the HSQC experiment.
