Through-space assignments in unlabeled nucleic acids
- Standard intraresidue sugar-to-base NOEs between anomeric H1' protons and base H6/H8 protons.
- Sequential assignment of imino and amino base resonances via imino-to-imino and imino-to-amino intraresidue and interstrand NOEs detected in NOESY experiments recorded in H2O.
- Stereospecific assignment of H5'/H5'' protons from NOESY if overlapping is not severe.
- Assignment of non-exchangeable base and sugar protons via NOE connectivities to amino and/or imino protons. Extensive overlapping can be resolved using hybrid experiments like 3D TOCSY-NOESY, TOCSY-NOESY or 3D NOESY-NOESY experiments.
