The 3D TOCSY-NOESY (TONO) experiment is specifically designed to obtain homonuclear 1H-1H NOE and J-coupling information in a single experiment. The experiment found widely application to study such effects in large unlabeled biomolecules having high overlapping resonances.REQUIREMENTS

The 3D TOCSY-NOESY pulse sequence ( 89JMR450-83 , 90JACS5024 , 91EJB807 , and 90JMR453-86 ) consists of a 2D TOCSY pulse sequence followed by a 2D NOESY pulse sequence.EXPERIMENTAL DETAILSSeveral modifications have been reported:
- Related 3D NOESY-TOCSY experiment has also been proposed by reversing the TOCSY and NOESY steps. Other analogs 3D experiments are easily obtained by changing one or both mixing process by others as, for instance, a ROESY scheme (3D TOCSY-ROESY experiment).
- Use of semiselective pulses to reduce the spectral widths in both non-detected dimensions ( 89JMR212-81 ).
- Application on proteins ( 90BIO1829 ) and nucleic acids ( 91JMR101-94 , 94JMRB134-103 ).
- 2D TOCSY-NOESY and doubly-selective 1D TOCSY-NOESY experiments have been also proposed to study smaller molecules.

The TOCSY-NOESY experiment affords a 3D spectrum in which several cross-peaks can be present:RELATED TOPICS
- Diagonal peaks.
- cross-peak due to single TOCSY transfer in the w2=w3 plane.
- cross-peak due to single NOESY transfer n the w1=w2 plane.
- cross-peak due to TOCSY and NOESY transfers.
